Nice ink!
I like the difference between thick and thin.
Try to make your lines sharper and smoother.
Jimmy's foot, for example, is kinda wobbly (is that a word? well I think you know what I mean right :)).
If you drew a line with the brush, you can edit it with the pen & pencil tool in Illustrator.
Mostly the less anchorpoints, the better.

Very nice. Have you tried changing thickness on selected lines after they're drawn? or it easier to redraw them?

I wish John or someone would do a video tutorial, inking one of his drawings (or maybe share their preferred brush settings). A comment on his blog said that "Manga Studio" has a canvas rotate option but I have yet to try it.

Thanks for the comments. I just started inking with Illustrator so I'm still trying to figure out what works for me.

For a few of my inks I tried drawing everything with the same brush then went back and changed the brush size, but sometimes the ends of the lines would pop out if you made it too big, or would leave gaps if you shrunk the line too much.

Right now I'll change the brush size when I draw, then when I'm done I'll go back and tweak the line width even more, adjusting the end points if need be, and if it doesn't work I'll just go ahead and redraw the line.

So you don't need to change the brush-size all the time:
You can make your own brushes in Illustrator.
Make a caligraphic brush and set the diameter to something you like, then select presure and set variation to the same as diameter.
With this you can make as much brushes as you like :).
I started with 2 brushes, but now I use 4.
